Values for Tm
Home > DNA Amplification > Overview > Primer Design > Values for Tm
An important consideration in primer design is to ensure that the 3′ and 5′ primers do not have very different values for Tm. Primers with a greater than 5°C difference in Tm may result in inadvertent preferential amplification of the most efficiently primed product strand. Large differences in Tm can lead to reduction in yield or even no amplification.
